Regulation of hepatic connexins during cholestasis: possible involvement of Kupffer cells and inflammatory mediators
نویسندگان
چکیده
Hepatocyte gap junction proteins, connexins (Cxs) 26 and 32, are down regulated during obstructive cholestasis (OC) and lipopolysaccharide hepatocellular cholestasis (LPS-HC). We investigated rat hepatic Cxs during ethynylestradiol hepatocellular cholestasis (EE-HC) and choledochocaval fistula (CCF) and compared them with OC and LPS-HC. Levels (immunoblotting) and cellular distribution (immunofluorescence) of Cxs 26, 32 and 43, as well as macrophage infiltration, were studied in livers of rats under each condition.culture with Kupffer cells. Thus, hepatocyte gap junctions are down regulated in forms of cholestasis associated with inflammation, and reduced intercellular communication might be induced in part by proinflammatory mediators.

Recent advances in clinical protocols have improved the outcomes of pancreatic islet transplantation (PIT), yet PIT recipients typically require pancreatic islet grafts derived from multiple donors to achieve insulin independence. This along with experimental models of syngeneic PIT, showing that up to 60% of pancreatic islet tissue undergoes apoptosis within the first several days post-transpl...
متن کاملThe functional role of microRNAs in alcoholic liver injury
The function of microRNAs (miRNAs) during alcoholic liver disease (ALD) has recently become of great interest in biological research. Studies have shown that ALD associated miRNAs play a crucial role in the regulation of liver-inflammatory agents such as tumour necrosis factor-alpha (TNF-α), one of the key inflammatory agents responsible for liver fibrosis (liver scarring) and the critical cont...
متن کاملPrecision-cut liver slices in culture as a tool to assess the physiological involvement of Kupffer cells in hepatic metabolism
Introduction Hepatic macrophages have the capacity to secrete a tremendous array of molecules, which can be divided into 3 categories – cytokines (TNF-alpha), lipid mediators (prostaglandins PGE2) and reactive intermediates (NO·) – in response to stimulus, such as lipopolysaccharides (LPS) [1,2]. Such mediators are capable to modulate both the metabolism and the integrity of hepatocytes in vitr...
متن کاملPossible Involvement of Liver Resident Macrophages (Kupffer Cells) in the Pathogenesis of Both Intrahepatic and Extrahepatic Inflammation
Liver resident macrophages designated Kupffer cells (KCs) form the largest subpopulation of tissue macrophages. KCs are involved in the pathogenesis of liver inflammation. However, the role of KCs in the systemic inflammation is still elusive. In this study, we examined whether KCs are involved in not only intrahepatic inflammation but also extrahepatic systemic inflammation. Administration of ...
